ANALISIS HOPANOID PADA SINORHIZOBIUM FREDII

ABSTRAK

Analisis kandungan hopanoid dari beberapa bakteri merupakan langkah eksperimen untuk membuktikan dugaan bahwa hopanoid dapat dijadikan marker dalam kemotaksonomi. Keberadaan hopanoid dalam Sinorhizobium fredii yaitu suatu bakteri fiksasi nitrogen yang hidup dalam tanah perlu ditelitiuntuk mencari hubungan kekerabatan antara bakteri penyubur tanah. Kandungan hopanoid kompleks dalam sel kering diekstrak dengan pelarut kloroformmethanol 21 vv . Oksidasi dan reduksi hopanoid menggunakan pelarut asam periodat dan natrium boro hidrida. Asetilasi hopanoid menggunakan pelarut piridin asetat anhidrat 11 vv .Langkah akhir hopanoid terasetilasi dideteksi menggunakan Kromatograf Gas- Spektroskopi Massa KG-SM. Keberadaan fragmen-fragmen spesifik mz 73 191 295 dan 369 membuktikan bahwa Sinorhizobium fredii mengandung hopanoid bis- homohopan 31 32 - diol Asetat


ABSTRACT

Analisis of hopanoid content of few bacterias is away to prove that hopanoid can beused as chemotacsonomy marker. The presence of hopanoid in Sinorhizobium fredii a bacteria nitrogen fixation live in the land has been studied in look for relation among nitrogen fixation bacteria. Complek hopanoid content in the bacterias dry cell were extracted by chloroform methanol 21 vv . Oxidation and reduction of hopanoid complek using periodat acid Natrium borohidrida. Acethylated of hopanoid using solvent piridin anhidrat acetat 11 vv . End step of hopanoid detectioned using Gas Cromatografi Massa Spectroscopi GC-MS . Specific hopanoid fragmen-fragmen existence mz 73 191 295 and 369 evidence that Sinorhizobium fredii content hopanoid bis- homohopan 3132 - diol Acetat
