EXTRACTION OF INULIN FROM ONION Allium cepa AND ITS BIOACTIVITY TEST AS SINBIOTIC COMPONENT WITH Lactobacillus casei strain BIO 251 AGAINTS BACTERIA CAUSES DIARRHEA

ABSTRAK

Inulin merupakan salah satu prebiotik yang berupa suatu polimer dari unitunit fruktosa yang berikatan glikosida pada rantai 2-1 dan diakhiri oleh unit glukosa. Inulin dapat diperoleh melalui ekstraksi dari bahan alam. Pada penelitian ini inulin diekstraksi dari umbi dahlia Dahlia pinnata menghasilkan randemen 399 dan bawang merah Allium cepa dengan randemen 020. Inulin dari umbi dahlia memiliki gula bebas total sebesar 39 kelarutan dalam air dingin sebesar 70 daya serap air 114 dan hasil analisis KLT menunjukkan Rf 050. Inulin dari bawang merah memiliki gula bebas total sebesar 25 kelarutan dalam air dingin sebesar 105 daya serap air 111 dan hasil analisis KLT menunjukkan Rf 049. Baik ekstrak inulin umbi dahlia maupun bawang merah pada konsentrasi 4378 ppm dapat meningkatkan pertumbuhan probiotik Lactobacillus casei strain BIO 251 masing-masing sebesar 526 dan 481. Sinbiotik yang mengandung ekstrak inulin baik dari umbi dahlia maupun bawang merah dengan konsentrasi minimal 4348 ppm dapat menghambat pertumbuhan Salmonella thypi yang merupakan salah satu bakteri penyebab diare masingmasing sampai 926 dan 914.


ABSTRACT

Inulin is prebiotic polymers mainly comprising of fructose units in wich are bond by a 2-1 glycosidic link and typically have a terminal glucose. Inulin can be extracted from natural source. At this research inulins extraction from dahlia root produces yield of inulin 3.99 and onion Allium cepa with yield of inulin 0.2. Extract of inulin from dahlia roots has 3.9 reduce sugar 7.0 cold water solubility 11.4 water binding capacity and result of TLC analysis shows retention time Rf 0.50. Extract of inulin from onion has 2.5 reduce sugar 10.5 cold water solubility 11.1 water binding capacity and result of TLC analysis shows Rf 0.49. Extract of inulin at concentration 437.8 ppm both inulin from dahlia root and onion are able to influent Lactobacillus casei strain Bio 251 there are 52.6 and 48.1. Synbiotic that containing extract of inulin both inulin from dahlia root and from onion with minimum concentration 434.8 ppm can be inhibited Salmonella thypi that is one of bacteria causes diarrhea which values 94.6 and 91.4 respectively.
