ABSTRAK

Enzim ligninolitik yang diisolasi dari jamur pelapuk kayu putih Phanerochaete chrysosporium dapat digunakan untuk mendegradasi lignin. Pemisahan enzim ligninolitik dilakukan dengan menggunakan metode pengendapan dengan menambahkan garam ammonium sulfat. Aktivitas enzim ligninolitik didefinisikan sebagai jumlah lignin yang terdegradasi setelah diinkubasi selama 1 hari. Uji aktivitas katalitik menunjukkan adanya aktivitas enzim ligninolitik terbesar dengan penambahan garam ammonium sulfat sebesar 50 vv dan mempunyai aktivitas sebesar 0.4 x 10-3 mgg.menit. Enzim yang ligninolitik yang mampu mendegradasi lignin selanjutnya dikarakterisasi dengan variasi konsentrasi substrat 100 hingga 600 ppm dengan interval 100 ppm vv. Aktivitas enzim ligninolitik ditentukan kandungan sisa substratnya lignin dengan menggunakan spektrofotometer UV-Vis. Hasil uji degradasi lignin diperoleh enzim ligninolitik hasil isolasi mempunyai aktivitas tertinggi 67.13 x 10-3 mgg.menit dengan konsentrasi substrat 600 ppm.


ABSTRACT

The ligninolytic enzyme which was obtained from isolated white-rot fungus P.chrysosporium has been used to reduce the lignin. The enzyme separation was done with precipitation method by adding the salt of ammonium sulphate. Activity of enzyme ligninolytic is defined as a number of lignin degradated after incubation for 1 day ammonium sulphate as much as 50 vv and the activity of ligninolityc enzyme was 123. Therefore this positive enzyme was characterited with variation of substrate concentration 100-600 ppm with interval 100 ppm vv. Activity ofl igninolityc enzyme was determined by spectrophotometer UV-Vis technique. The result of lignin degradation test showed the highest activity of isolated ligninolytic enzyme is 4457 with substrate 600 ppm
