ABSTRAK

Lignin merupakan suatu kompleks polimer aromatik dan mempunyai berat molekul sangat tinggi. Senyawa aromatik berantai panjang tersebut sulit terurai dan bersifat toksik merupakan hasil proses delignifikasi limbah industri kertas. Lignin dapat didegradasi oleh enzim ligninolitik amobil yang diisolasi dari jamur Phanerochaete chrysosporium. Pada penelitian ini dilakukan degradasi lignin menggunakan enzim ligninolitik yang telah diamobilisasi menggunakan agarose dengan variasi konsentrasi substrat lignin antara 100 600 ppm selama 4 hari dengan penyamplingan tiap 2 hari. Besarnya konsentrasi lignin yang terdegradasi oleh enzim ligninolitik amobil ditentukan dengan mengukur absorbansinya menggunakan spektrofotometer UV. Berdasarkan penelitian yang telah dilakukan diperoleh bahwa konsentrasi substrat dan waktu inkubasi dapat mempengaruhi banyaknya lignin yang terdegradasi. Semakin tinggi konsentrasi substrat maka semakin besar konsentrasi lignin yang terdegradasi tetapi semakin lama waktu inkubasi maka aktivitasnya akan menurun. Aktivitas enzim ligninolitik amobil tertinggi pada konsentrasi lignin 600 ppm sebesar 346.5 unit dengan waktu inkubasi 2 hari


ABSTRACT

Lignin is a complex aromatic polymer and has a very high molecular weight. This long chained aromatic compound that difficult to decompose and toxic is the result of delignification process of paper industrial waste. This compound can be degraded with ligninolitik enzyme amobil that isolated from Phanerochaete chrysosporium In this research lignin degradation process was performed. The process was using ligninotik that has been amobilizised using agarose with the variation of the substrate concentration was between 100-600 ppm within 4 days period with every two days of sampling . The amount of lignin concentration that degraded by the amobile ligninolitic enzyme was determine by measuring the absorbent using spectrophotometer UV. Based on the research the result shows that the substrate concentration and incubation period can have an impact to the amount of degraded lignin. The higher the substance concentration is the higher of the concentration of lignin that degraded. However the longer the incubation period is the lower of their activities. The highest activities of amobile ligninolitik enzyme at 600 ppm is 346.5 unit with the incubation period of 2 days.
