SELEKSI ISOLAT KAPANG TANAH WONOREJO PENGHASIL ENZIM LIPASE

SELECTION OF WONOREJOS SOIL MOLD ISOLATE FOR LIPASE ENZYME PRODUCER

ABSTRAK

Kemampuan 37 isolat kapang tanah Wonorejo Surabaya dalam menghasilkan enzim lipase serta aktivitas enzimatiknya telah diteliti. Minyak zaitun dan rhodamine B yang ditambahkan ke dalam medium basal digunakan sebagai medium seleksi. Berdasarkan parameter terbentuknya zona berpendar dapat terlihat bahwa dari 37 isolat yang diseleksi terdapat 23 isolat yang mampu menghasilkan enzim lipase. Dari 23 isolat kapang tersebut terdapat tujuh isolat yang dapat menghasilkan zona berpendar dengan intensitas pendaran yang tinggi yaitu Mortiriella sp. Penicillium sp.1 Fusarium sp. Gliomastix sp.1 Gliomastix sp.2 Mycelia sterilia Unk1 dan Mycelia sterilia Unk2. Uji aktivitas lipase dilakukan pada ketujuh isolat tersebut serta Aspergillus niger. Aspergillus niger digunakan juga pada uji aktivitas karena pada studi literatur menunjukkan bahwa isolat Aspergillus niger menghasilkan aktivitas lipase yang tinggi. Isolat dengan aktivitas lipase tertinggi pada medium cair adalah Mortiriella sp. 00077 UmL. Isolat dengan aktivitas lipase dari nilai yang terbesar hingga terkecil selanjutnya adalah Gliomastix sp.1 00073 UmL Aspergillus niger 00063 UmL Mycelia sterilia Unk2 00058 UmL Mycelia sterilia Unk1 00043 UmL Penicillium sp.1 00037 UmL Gliomastix sp.2 00034 UmL dan kemudian Fusarium sp. 00032 UmL.


ABSTRACT

The ability of isolates of the soil mold in Wonorejo Surabaya produces the lipase enzymes and the enzimatic activities have been researched. The selection medium used the basal mediums which is contain olive oil and rhodamine B. Based on the formation parameters 23 isolates amounts of the isolate in this research is 37 isolates can be seen at the flourescent zone and they produced the lipase enzymes. Seven of those 23 isolates of the mold produce a flourescent zone with a high luminescence intensity ie Mortiriella sp. Penicillium sp.1 Fusarium sp. Gliomastix sp.1 Gliomastix sp.2 Mycelia sterilia Unk1 and Mycelia sterilia Unk2. Lipase activity assay was performed on those seven isolates and Aspergillus niger. This activity assay used the Aspergillus niger due to the study of the literature showed that Aspergillus niger produces high lipase activity. Mortiriella sp. 00077 UmL is the isolate that has the highest lipase activity in the liquid medium. The next level of each isolates lipase activity are Gliomastix sp.1 00073 UmL Aspergillus niger 00063 UmL Mycelia sterilia Unk2 00058 UmL Mycelia sterilia Unk1 00043 UmL Penicillium sp.1 00037 UmL Gliomastix sp.2 00034 UmL and Fusarium sp. 00032 UmL.
