SCREENING OF PHOSPHATE SOLUBILIZING AND ORGANIC SUBSTRATE DEGRADING YEAST FROM POTERAN ISLAND MADURA

ABSTRAK

Peningkatan potensi pulau Poteran Madura dapat dilakukan melalui perbaikan kualitas pada lahan pertanian yakni dengan memanfaatkan khamir sebagai agen biofertilizer Penelitian ini bertujuan untuk menginventarisasi mengidentifikasi khamir dari pulau Poteran Madura yang berperan melarutkan fosfat dan mendegradasi bahan organik. Identifikasi khamir hingga tingkat genus berdasarkan karakteristik morfologi makroskopis dan mikroskopis reproduksi seksual serta uji fisiologis. Uji potensi dalam degradasi selulosa dengan CMC agar degradasi proteolitik dengan skim milk agar degradasi lignin pada media LAC dan pelarut fosfat pada media Pykovskaya. Kemampuan khamir ditunjukkan dari zona bening yang terbentuk disekitar koloni. Hasil penelitian diketahui isolat yang berpotensi pelarut fosfat diduga genus Candida dan Saccharomyces dengan indeks pelarut fosfat tertinggi sebesar 0.65 oleh genus Saccharomyces. Khamir yang berpotensi selulolitik adalah Candida Filobasidium Trichosporon dan Saccharomyces dengan IS tertinggi sebesar 4.17 oleh genus Candida. Khamir yang berpotensi proteinase adalah Pichia dan Trichosporon dengan IP tertinggi sebesar 0.09 oleh genus Pichia dan semua genus yang ditemukan tidak berpotensi mendegradasi lignin.


ABSTRACT

Increasing of Poteran island Madura potency can be done through the improvement of agricultural quality by using yeast as a biofertilizer agent. This study aims to inventory identify yeasts from Poteran island Madura which plays a role in solubilizing phosphate and degrade organic substrate. Yeast isolates were identified to the genus based on macroscopic and microscopic morphological characteristics sexual reproduction and physiological tests. The potential degradation test of cellulose on CMC media proteolytic degradation with skim milk agar lignin degradation in LAC and solubilizing phosphate in Pykovskaya agar. The potency of yeast was showed from the clean zone. The results of this study are known that the potential of solubilizing phosphate were Candida and Saccharomyces with the highest index value solubilizing approxiamtely at 0.65 of the Saccharomyces. Cellulolytic isolates were Candida Filobasidium Trichosporon and Saccharomyces with the highest index value cellulolytic approximately at 4.17 of the Candida. Isolates that have the potential proteolytic were Trichosporon and Pichia with proteolytic index IP approximately at 0.09 of the Pichia and all of the genus were not obtained as a potentially degrade lignin.
