PEMBENTUKAN KOMPLEKS INKLUSI KETOPROFEN-946-CYCLODEXTRIN DENGAN MENGGUNAKAN KARBONDIOKSIDA SUPERKRITIS

FORMATION OF KETOPROFEN- 914-CYCLODEXTRIN INCLUSION COMPLEXES USING SUPERCRITICAL CARBONDIOXIDE

ABSTRAK

Perkembangan ilmu pengetahuan dan teknologi yang pesat khususnya bidang farmasi telah menghasilkan perubahan yang signifikan dalam teknologi sediaan farmasi khususnya obatobatan. Obat yang memiliki kelarutan rendah proses absorbsinya ditentukan oleh tahap disolusi rate limiting step. Untuk meningkatkan laju disolusi dapat dilakukan dengan cara pembentukan kompleks inklusi obat dalam 946-cyclodextrin dengan Teknologi Supercritical Fluid SCF. Salah satu cara meningkatkan kompleksasi dari suatu kompleks inklusi obat dalam 946-cyclodextrin dapat dilakukan dengan menambahkan air pada campuran obat- 946-cyclodextrin. Tujuan dari penelitian ini adalah memperoleh kompleks inklusi ketoprofen- 946-cyclodextrin dengan menggunakan teknologi supercritical fluid mempelajari pengaruh perbedaan tekanan saturation vessel dan penambahan air terhadap kompleks inklusi ketoprofen- 946-cyclodextrin. Penelitian ini diawali dengan membuat campuran ketoprofen- 946- cyclodextrin-air yang diaduk selama 10 menit dengan komposisi 15 lalu disaturasi pada suhu 50 oC dan variasi tekanan 80-240 bar selama 2 jam disertai pengadukan kemudian dikeringkan. Karakterisasi kompleks inklusi dilakukan dengan analisa Differential Scanning Calorimetry DSC Scanning Electron Microscopy SEM Drug Release dan Drug Loading. Dari penelitian ini dapat diperoleh kesimpulan kompleks inklusi ketoprofen- 946 Cyclodextrin dapat dihasilkan menggunakan karbondioksida superkritis dengan kompleksasi antara 829- 100. Uji drug loading dari kompleks inklusi pada tekanan 80 bar cenderung memenuhi syarat Farmakope Indonesia dengan rentang 8729-10452. Uji drug loading pada penambahan 40 air cenderung memenuhi syarat Farmakope Indonesia dengan rentang 7692-10034. Uji drug release komplek inklusi tekanan 80 120 bar dan pada penambahan air 60 dan 80 memiliki laju disolusi yang lebih cepat dibandingkan dengan ketoprofen murni.


ABSTRACT

Development of sciences and technology is growing especially in pharmaceuticals has resulted in significant changes in the technology of pharmaceutical preparations such as drugs. The absorption process of drugs with low solubility determined by dissolution step rate limiting step. To improve the dissolution rate can be done by means of the formation of inclusion complexes 946-cyclodextrin drug with Supercritical Fluid SCF. One way to improve the complexation of an inclusion complex of 946 - cyclodextrin in the drug can be made by adding water to the mixture of 946 - cyclodextrin - drug . The purpose of this study was to obtain inclusion complexes of ketoprofen - 946 - cyclodextrin using supercritical fluid technology to study the effect of different saturation pressure vessel and the addition of water to the inclusion complex of ketoprofen - 946 - cyclodextrin. This study begins with a mixture of ketoprofen 946-cyclodextrin ratio 15 ww and added water by manually blended using mortar for about 10 minutes. Then be saturated at 50 oC with variations of the pressure of supercritical carbon dioxide for about 2 hours and mixed continuously then soon be dried.. Characterization of inclusion complexes performed with analysis Differential scanning calorimetry DSC Scanning Electron Microscopy SEM Drug Release and Drug Loading . From this study can be concluded ketoprofen inclusion complex 946 - Cyclodextrin can be produced using supercritical carbon dioxide by complexation between 82.9 to 100 . Drug loading analysis of complex inclusion at 80 bar pressure tends qualified Farmakope Indonesias with a range of 87.29 to 104.52 . Drug loading analysis on the addition of 40 water tends qualified Farmakope Indonesias with a range of 76.92 to 100.34 . Drug release analysis of complex inclusion at pressure 80 120 bar and the addition of 60 80 water have a faster dissolution rate compared to pure ketoprofen.
