PENGARUH ION CU2 PADA MATRIKS AGAROSE TERHADAP KESTABILAN ENZIM BROMELAIN HASIL ISOLASI YANG AMOBIL

ABSTRAK

Amobilisasi enzim bromelain yang dihasilkan oleh buah nanas dilakukan pada gel agarose tanpa penambahan Cu2 dan dengan penambahan Cu2. Isolasi enzim bromelain dari buah nanas dilakukan dengan metode salting out menggunakan garam amonium sulfat 0 - 43S. Enzim bromelain yang diperoleh diamobilisasi pada gel agarose dengan variasi konsentrasi 1 2 3 menggunakan teknik amobilisasi adsorpsi. Penentuan konsentrasi sisa protein kasein yang digunakan sebagai substrat pada optimasi konsentrasi agarose dan pH dilakukan menggunakan metode Bradford pada panjang gelombang maksimum 590 nm. Hasil uji aktivitas enzimatik pada agarose 3 yang mengandung Cu2 adalah yang terbesar yaitu 2906 unitjam pada kondisi pH optimum 65 dan enzim bromelain amobil pada agarose 3 tanpa Cu2 hanya mampu mendegradasi kasein 270 unitjam.


ABSTRACT

Bromelain enzim which produced by pinneapple fruits is amobilized in agarose gel by adding Cu2 and without Cu2. The enzim is isolated by salting out method using amonium sulfate salts 0 - 43 . Then the enzim is amobilized in agarose gel by variating concentration 1 2 3 using adsorption amobilization technique. The rest of casein protein concentration which used as substrate is determined by optimizing agarose concentration and pH using Bradford method in maximum wave length 590 nm. The biggest enzimatic activity test is agarose 3 with Cu2 that 2906 unithour in the optimum pH 65. The amobilized bromelain enzim in agarose 3 without Cu2 degradates casein only 270 unithour.
