ABSTRAK

Proses pengelantangan bleaching process yaitu proses penghilangan sisa lignin pada industri pulp dan kertas menghadapi masalah yang serius terkait dengan penggunaan senyawa kimia khususnya klorin yang sangat polusif terhadap lingkungan. Metode alternatif yang ramah lingkungan mutlak diperlukan untuk mengatasi permasalahan ini yaitu dengan memanfaatkan enzim xylanase sebagai pengganti klorin pada proses bleaching. Penelitian ini bertujuan menghasilkan enzim xylanase yang merupakan enzim pendegradasi komponen hemiselulose di dalam kayu. Karena hemiselulose merupakan perekat antara lignin dan selulose maka dengan terdegradasinya hemiselulose lignin yang terlepas dapat dengan mudah dihilangkan dengan hanya penambahan sedikit klorin atau bahkan tanpa menggunakan klorin sama sekali. Metodologi penelitian ini dimulai dengan mengembangkan Aspergillus niger dalam Potato Dextrose Agar PDA kemudian mempersiapkan substrat untuk media pertumbuhan. Setelah enzim diekstrak dari sel-sel mikroorganisme dilakukan purifikasi enzim menggunakan metode ammonium sulfate precipitation dilanjutkan dengan proses dialisis. Dari hasil percobaan diketahui dalam proses produksi enzim xylanase solid state fermentation SSF lebih baik digunakan sebagai metode dalam memproduksi enzim xylanase ini. Aktifitas enzim xylanase tertinggi adalah 3.1795 IUml dihasilkan oleh strain jamur Aspergillus niger dalam SSF dengan penambahan xylan sebagai inducer. Purifikasi enzim dengan ammonium sulfate precipitation dan proses dialysis masih belum mampu meningkatkan aktifitas xylanase yang dihasilkan namun mampu menurunkan aktifitas spesifik enzim selulase.


ABSTRACT

Bleaching process the removal process rest of lignine in pulp and paper industry faces a serious problem with the use of chemical compound specially chlorine that very polutive to the environment. Alternative methods that environmentally friendly absolutely needed to overcome this problem by exploiting xylanase enzymes as chlorine subtitutes in bleaching process. This experiment is aimed to yield xylanase enzymes the degrading hemicellulose enzymes in woods. Since the hemicellulose is a glue between lignine and cellulose by hemicellulose degradation lignine will be easier to removed by adding a little chlorine or nothing at all. This experiment methods is begin by growing Aspergillus niger in Potato Dextrose Agar PDA prepare the substrate for growing medium. After extraction enzymes from microorganism cells purification is done by ammonium sulfate precipitation and dialysis. From the result of experiment we know that in production xylanase enzymes solid state fermentation SSF is better use than submerge fermentation. The highest activity of xylanase enzymes is 3.1795 IUml yielding by Aspergillus niger in SSF with addition of xylan as inducer. Purification of xylanase enzymes with ammonium sulfate precipitation dan dialysis have not yet increased the spesific activity of xylanase activity but could decreasing the cellulase activity.
