ABSTRAK

Penelitian dilakukan untuk mengetahui aktivitas enzim ligninolitik yang diproduksi oleh jamur pembusuk kayu putih Trametes versicolor. Aktivitas enzim ligninolitik diukur dengan menggunakan substrat lignin yang diisolasi dari kayu lamtoro dengan metode pulp kraft. Untuk merangsang produksi enzim ligninolitik dari jamur T. versicolor maka jamur tersebut diadaptasikan terlebih dulu dalam media pertumbuhan yang mengandung lignin. Karena enzim ligninolitik diproduksi secara ekstraseluler maka hasilnya terdapat dalam media cair hasil fermentasi. Untuk mengisolasi enzim ini dilakukan dengan metode pengendapan menggunakan larutan gradien garam amonium sulfat pada kejenuhan 10-50. Enzim ligninolitik dengan kemurnian yang paling tinggi diperoleh pada pengendapan dengan kejenuhan 20-30 00385 mgg.min. Pengaruh konsentrasi terhadap aktivitas enzim yang terbesar diperoleh pada konsentrasi lignin 500 ppm aktivitas enzim ligninolitik 01414 mgg.min.


ABSTRACT

This experiment was done to know the activity of extraceluler ligninolytic enzyme which is produced by white rot fungi T. versicolor. The activity of ligninolytic enzyme was measured by using lignin substrat which is isolated from Lamtoro wood by using kraft pulp methode. This fungi must be adapted first on a slant culture that containing lignin. The enzyme was obtained by amonium sulphate precipitation methode at 10-50 of saturation. Enzyme precipitated by 20-30 of saturation amonium sulphate represent as a highest purified enzyme 00385 mgg.min. The highest activity of ligninolytic enzyme 01414 mgg.min was achieved at substrat concentration of 500 ppm
