ISOLATION AND IDENTIFICATION OF AEROBIC BACTERIA WHICH DEGRADE LIPID IN ORGANIC WASTE AT TPS TEMPORARY DISPOSAL SITE BENTENG SURABAYA

ABSTRAK

Sampah organik adalah sampah yang sebagian besar tersusun oleh senyawa organik Sampah organik memiliki kandungan karbohidrat protein dan lemak yang tinggi. Bakteri memiliki kemampuan untuk menguraikan bahan organik. Lemak disebut juga lipid adalah suatu zat yang kaya akan energi berfungsi sebagai sumber energi yang utama untuk proses metabolisme. Tujuan dari penelilitian ini adalah mengisolasi dan mengidentifikasi bakteri aerob pendegradasi lemak pada sampah organik di TPS Benteng. Isolasi menggunakan medium selektif lemak Isolat diIdentifikasi menggunakan uji biokimia pewarnaan Gram pewarnaan spora dan uji biokim dengan uji fermentasi karbohidrat laktosa glukosa dan sukrosa oksidase katalase motilitas IMViC Indol Methyl Red Voges-Proskauer Citrat dan TSIA Triple Sugar Iron Agar. Hasil isolasi ditemukan 54 isolat bakteri yang dapat Digolongkan kedalam 5 genus yaitu Serratia Neisseria Staphylococcus Streptococcus dan Lactobacillus.


ABSTRACT

Organic waste is waste that composed by organic matter. Organic waste composed by a lot of carbohydrate protein and lipid. Bacteria have an ability to degrade the organic matter. Lipid have a lot of energy it can be a source of energy for the metabolism progress. The purpose of this research is finding out the kind of aerobic bacteria in organic garbage that degrade the lipid matter at TPS Benteng. The lipid selective media used isolation. Isolate was identified with Gram stain spore stain and biochemical test with carbohydrate fermentation test lactose glucose and sucrose oxidase catalase motility IMViC Indol Methyl Red Voges-Proskauer Citrat and TSIA Triple Sugar Iron Agar. The result of the isolation founded 54 isolates bacteria that can be grouped in five Genus those are Serratia Neisseria Staphylococcus Streptococcus and Lactobacillus.
