ABSTRAK

Lumpur Lapindo mempunyai salinitas tinggi dan bertekstur liat berdebu. Bibit Crotalaria striata yang ditanam pada media lumpur LapindoBokashi dapat bertahan sampai dengan 6 hari. Penelitian bertujuan untuk mengetahui profil protein Crotalaria striata yang ditanam pada media lumpur LapindoBokashi. Bokashi digunakan sebagai pupuk dasar ditambahkan pada 50 gram lumpur Lapindo dengan komposisi 0 50 100 dan 150 gram. Kontrol digunakan tanah habitat asli. Penelitian dilakukan mulai bulan Mei sampai dengan Juli 2008. Pemeliharaan tanaman di dalam green house. Analisis profil protein SDS-PAGE 15 dilakukan di Laboratorium Institute of Tropical Diesease Surabaya. Perlakuan kontrol dan Bokashi 150 gram membentuk 9 pita protein sedangkan perlakuan Bokashi 100 50 dan 0 gram membentuk 12 pita protein. Adanya tambahan 3 pita protein yang berbeda dengan kontrol diasumsikan tanaman membentuk protein yang spesifik pada cekaman kekeringan senyawa kompatibel yaitu protein dengan berat molekul 30 kDa 11875 kDa dan 105 kDa. Cekaman kekeringan juga berpengaruh terhadap morfologi Crotalaria striata pertambahan tinggi tanaman akan meningkat seiring dengan jumlah penambahan Bokashi dan jumlah daun akan berkurang pada perlakuan tanpa Bokashi.


ABSTRACT

Lapindo mud has high salinity and dusty rubbery texture. The seedsof Crotalaria striata are planted in Lapindo mud and Bokashi can hold out until six day. The purpose of this experiment are to know the profile protein of Crotalaria striata that planted in Lapindo mud and Bokashi. Bokashi is used basic fertilizer that added on 50 gram of Lapindo mud the compositions are 0 50 100 and 150 gram. The pure sand of the habitat is used for control. This experiment was held since May until July 2008. Taking care the plant in green house. Analysis the profile protein SDS PAGE 15 was held in Institut of Tropical Disease Laboratory Surabaya. Control and Bokashi 150 gram treatment to form 9 bands whereas bokashi 100 50 and 0 gram treatments to form 12 bands protein. There is added 3 bands protein that the specific protein in water stress compatible compound is protein with molecule weight 30 kDa 11875 kDa and 105 kDa. Water stress also infected to the morphology of Crotalaria striata added of amount of Bokashi increase the high of the plant and amount of leafs will decrease in treatment without Bokashi.
