PRODUKSI ETANOL MENGGUNAKAN MUTAN ZYMOMONAS MOBILIS YANG DIMUTASI DENGAN HYDROXYLAMIN

ETHANOL PRODUCTION USING Zymomonas mobilis MUTANT MODIFIED BY HYDROXILAMINE MUTAGEN

ABSTRAK

Proses mutasi yang dilakukan pada penelitian ini merupakan upaya untuk meningkatkan produktivitas Zymomonas mobilis. Mutasi dilakukan dengan menggunakan mutagen hydroxylamine. Mutant dikarakterisasi dengan media asam pH 4 45 5 6 dan dibandingkan dengan pH normal bentuk sel dan pola pertumbuhan. Mutant Zymomonas mobilis kemudian disebut Zymomonas mobilis A3. Zymomonas mobilis A3 menghasilkan etanol tertinggi 048 g etanol g glukosa atau 9546 etanol teoritis pada pH awal 45. Perubahan bentuk sel Zymomonas mobilis terjadi setelah proses mutasibentuk sel batang bercemeti kecil dan aktif bergerak menjadi sedikit lebih besar dengan gerakan lebih pelan. Pola pertumbuhan sel menjadi lebih singkat dengan fasa adaptasi selama 3 jam puncak fasa eksponensial terjadi pada 14 jam dan fasa stasioner terjadi pada jam ke 14-16. Zymomonas mobilis A3 difermentasikan selama 40 jam dan rendemen diukur tiap jam. Produktifitas etanol tertinggi didapatkan pada fermentasi selama 35 jam dengan nilai 1301 gLjam dan jumlah biomassa 575 x 1010sel mL.


ABSTRACT

Mutation of Zymomonas mobilis is an effort to increase productivity of Zymomonas mobilis in learn of alcohol production. Mutation was induced by hydroxylamine mutagen. Mutant was selected with acid medium pH 4 45 5 6 and characterized with cell morfology and growth rate pattern. Zymomonas mobilis mutant obtain was then called Zymomonas mobilis A3. The highest yield of Zymomonas mobilis A3 048 g etanol g glucose or 9546 theoritical etanol in ethanol fermentation in form occur at pH 45. Zymomonas mobilis morfology change after mutation process. Zymomonas mobilis A3 cell are bigger than those of Zymomonas mobilis wild type. In addition they are less mobile than Zymomonas mobilis wild type . Required time for adaptation and exponential phase are lower than the time required for Zymomonas mobilis wild type.. Fermentation Zymomonas mobilis A3 was conducted for 40 hours and the result was measured every hour. The highest productivity of etanol found after 35 hours with value 1301 gLh and count of biomass 575 x 1010cell mL.
