PRODUCTION OF ACTIVE PHARMACEUTICAL INGREDIENTS API - POLYMER COMPOSITE MICROPARTICLES BY USING SUPERCRITICAL CARBONDIOXIDE

ABSTRAK

Ketoprofen asam RS-2-3-benzoylphenyl-propionat termasuk jenis non-steroidal anti-inflammatory drugs NSAID yang dikenal sebagai anti inflammasi antipiretik dan anlgesik. Ketoprofen memiliki kelarutan yang rendah dalam air. Untuk memperbaiki laju kelarutannya dilakukan dengan cara mikronisasi dan memformulasikan Active Pharmaceutical Ingredients API dengan polimer sebagai drug excipients. Penelitian ini bertujuan memperoleh mikropartikel komposit Active Pharmaceutical Ingredients API-polimer dengan teknologi supercritical anti solvent SAS dan particles from gas-saturated solutions PGSS. Pada metode SAS dipelajari pengaruh tekanan presipitator temperatur kontak dan konsentrasi awal larutan obat-polimer. Sedangkan pada metode PGSS dipelajari pengaruh oleh tekanan dan temperatur saturation chamber dan komposisi obat-polimer. Pada metode SAS ketoprofen dan polimer polietilen glikol PEG dilarutkan dalam etanol-aseton kemudian dikontakkan dengan fluida superkritis dan diekspansikan ke dalam presipitator melalui nozzle. Pada metode PGSS ketoprofen dan polietilen glikol PEG dilelehkan terlebih dahulu dengan fluida superkritis kemudian diekspansikan melalui nozzle. Mikropartikel yang terbentuk dianalisa dengan menggunakan SEM Scanning Electron Microscopy untuk mengetahui morfologi ukuran partikel dan distribusi ukuran partikel yang dihasilkan. Uji drug loading dan drug release dilakukan dengan spektrofotometer UV-Vis. Pada penelitian ini diperoleh hasil yaitu diameter rata-rata partikel sebesar 92 148 m untuk metode SAS dan 77 114 m untuk metode PGSS. Hasil uji drug loading menunjukkan efisiensi drug loading untuk mikropartikel dari metode SAS sebesar 8146 - 10090 dan 7479 - 9984 untuk metode PGSS. Uji drug release menunjukkan bahwa komposit ketoprofen-PEG memiliki laju disolusi lebih tinggi dibandingkan dengan ketoprofen tanpa proses mikronisasi. ABSTRACT

Ketoprofen RS-2-3-benzoylphenyl-propionic acid is a non-steroidal antiinflammatory drug with well-known anti inflammatory antipyretic and analgesic properties. Solubility of ketoprofen is low. To improve the rate of dissolution was performed by micronization and make a formulation of a Active Pharmaceutical Ingredients API and polymer as drug excipients. The purpose of this research is to get Active Pharmaceutical Ingredients-polymer composite microparticles by Supercritical anti-solvent technology SAS and particles from gas-saturated solutions PGSS. In the SAS method is studied the influence of precipitator pressure contact temperature and initial concentration of solution drugs-polymer. While in PGSS method is studied the influence of pressure and temperature of saturation chamber and the drugs-polymer compositions. In the SAS method ketoprofen and the polyethylene glycol PEG are dissolved in acetone-ethanol then contacted with supercritical fluids and sprayed into precipitator through nozzle. In PGSS Method ketoprofen and PEG are melted with supercritical fluid and then sprayed through a nozzle. Microparticles are analyzed by using SEM Scanning Electron Microscope to determine morphology particle size and particle size distribution. Drug loading and drug release test have done by using UV-Vis spectrophotometric. the results of this research obtained is the average diameter of particles of 92 to 148 956m for SAS method and from 77 to 114 956m for the PGSS method. Drug test showed that the drug loading efficiency for microparticles of SAS methode are 81.46 - 100.90 and 74.79 - 99.84 for the PGSS method. Drug release test showed that ketoprofen-PEG composite has a higher dissolution rate compared with ketoprofen without micronization. Keyword ketoprofen microparticles supercritical fluids SAS PGSS
