ABSTRAK

ABSTRAK Penelitian ini bertujuan untuk mempelajari pengembangan teknik immobilisasi enzim sebagai aplikasi biosensor dengan menggunakan metode sandwich dan pemerangkapan mempelajari pengaruh konsentrasi silika pada pembuatan sol-gel terhadap aktivitas enzim yang diimmobilisasi dan mempelajari pengaruh kondisi operasi pengukuran amperometrik terhadap aktivitas enzim yang diimobilisasi. Pada penelitian ini sol silika dibuat dengan menghidrat TEOS dalam air demin pelarut etanol dan katalis HCl sambil diaduk selama 15 jam dengan magnetic stirer dan kemudian diuapkan sebelum digunakan untuk mengimmobilisasi enzim. Ada dua macam teknik immobilisasi yang digunakan yaitu metode sandwich dan pemerangkapan. Biosensor yang telah jadi ini akan diuji aktivitas enzimnya terhadap respon arus ditinjau dari konsentrasi silika berat SiO2 potensial suhu dan pH. Berdasarkan uji amperometrik diperoleh respon arus yang baik adalah pada metode sandwich potensial 04 Volt konsentrasi SiO2 sebesar 108 loading enzim sebesar 35 Umm2 pH larutan 946-Dglukosa 7 dan suhu larutan 946-D-glukosa 27C


ABSTRACT

ABSTRACT The main objective of this research is to study the influence of enzyme immobilization techniques for its application as biosensor. The immobilization studied included a sandwich method and entrapment method. The effect of silica concentration in sol-gel preparation on the immobilized enzyme activity was studied. Moreoverthe stability of the immobilized enzyme against its environment such as pH and temperature was also investigated. The silica sol for immobilization enzim was prepared by hydrolysis of TEOS in ethanol uses HCl as catalyst. The ethanol was evaporated by flowing air before the sol was used for immobilization. There are two types of immobilization techniques used in this study the sandwich method and entrapment method.. The biosensor was tested by an a chrono amperometry method to evaluate their performance to detect glucose in solution. It has been shown that the sandwich design which is simpler in its fabrication. Such characteristic was obtained by the sandwich biosensor with enzyme loading of 35 umm2 at the concentration of 108 SiO2 at an applied potential of 0.4 V. The measurement was carried out at room temperature and pH 7.
