Pada penelitian ini telah dilakukan pengaruh komposisi etanol toluen pada permeabilisasi sel Zymomonas mobilis dan pengaruh konsentrasi substrat glukosa pada produksi etanol secara enzimatik. Sel Zymomonas mobilis diinokulasikan dalam medium cair selama 22 jam pada suhu ruang untuk memperoleh biomassa. Permeabilisasi dilakukan dengan menambahkan berbagai komposisi perbandingan etanol dan toluen ke dalam biomassa. Komposisi perbandingan etanol dan toluen yang digunakan adalah 1:4, 2:3, 3:2, 4:1. Pengukuran absorbansi larutan menggunakan spektrofotometer UV pada panjang gelombang 270 nm dilakukan untuk mengetahui keberhasilan proses permeabilisasi.. Untuk mengetahui kandungan etanolnya dilakukan uji secara kualitatif dan dapat dilihat dari perubahan konsentrasi substrat glukosa. Konsentrasi etanol tertinggi yang diasumsikan dari perubahan konsentrasi substrat glukosa terbesar dihasilkan oleh sel Zymomonas mobilis yang dipermeabilisasi menggunakan komposisi etanol toluen 1: 4 dan konsentrasi glukosa 10 % b/v.

In this research, the composition ethanol toluene effect and substrate concentration effect on permeabilized cell of Zymomonas mobilis have been investigated. Zymomonas mobilis cell were innoculated in liquid medium in 22 hours at room temperature to obtain biomass. Permeabilization was done by adding ethanol toluene into biomassa. The ratio of ethanol toluene compositions are 1:4, 2:3, 3:2, 4:1. Measurement filtrate absorbance using UV spektrophotometer at wavelength 270 nm was done to know the successfully of permeabilization process. To determine the ethanol, qualitatif methode has been done and also from the decreased of glucose concentration. The highest ethanol concentration assumed from the biggest dcreased glucose concentration received from Z. mobilis cell permeabilized with composition ethanol: toluen (1:4) and glucose concentration 10%.
