KEANEKARAGAMAN ANGGOTA GENUS AZOTOBACTER RESISTEN MERKURI MENGGUNAKAN PENDEKATAN TAKSONOMI NUMERIK FENETIK

DIVERSITY OF PHOSPHATE SOLUBILIZING BACTERIA PSB IN TALANGO AND POTERAN VILLAGE POTERAN ISLAND SUMENEP MADURA

ABSTRAK

Salah satu potensi Pulau Poteran Sumenep adalah bidang pertanian tetapi beberapa desa di Kecamatan Talango kondisi lahan kering dan kurang subur. Salah satunya disebakan oleh rendahnya P-tersedia padahal dilaporkan P total tanah Madura adalah sedang hingga tinggi. Oleh karena itu diperlukan adanya BPF. Penelitian ini bertujuan mengisolasi BPF tanah di Desa Talango dan Poteran agar diketahui keanekaragaman dan efisiensi pelarut fosfat bakteri tersebut. Isolasi bakteri dilakukan dengan metode pengenceran bertingkat dan metode sebar pada medium NA. Isolat murni diamati secara deskriptif makroskopis bentuk tepi elevasi dan warna koloni mikroskopis morfologi sel dan pewarnaan dan karakter biokimia berdasarkan panduan Bergeys Manual of Determinative serta diukur efisiensi pelarut fosfat dengan medium Pikovskaya. Terdapat 37 isolat bakteri dari Desa Talango dan Poteran yang berpotensi sebagai BPF dan kecenderungan masuk ke 12 genus berbeda yaitu Bacillus Corynebacterium Staphylococcus Vibrio Aeromonas Pseudomonas Escherichia Edwardsiella Providencia Proteus Shigella dan Yesrsinia. Ke-37 isolat menunjukkan rata-rata tingkat efisiensi pelarut fosfat sebesar 40 dimana genus Vibrio memiliki tingkat efisiensi paling tinggi yaitu 232 sehingga diasumsikan bakteri tersebut berpotensi sebagai biofertilizer.


ABSTRACT

Agriculture is one of the potential of Poteran Island Sumenep but in some village of Talango Subdistrict the soil condition are dry and poor nutrition. It is due to lack of available P whereas it had been reported that total P was moderat to high. This problem could be helped by PSB. This research purpose to isolate Phosphate Solubilizing Bacteria PSB in the Talango and Poteran village in order to know the diversity and phosphate solubilization efficiency of their bacteria. Bacterial Isolation was carried out with serial dilution method and spread plate method on the NA medium. Pure isolate was observed in macroscopic character shape margin elevation and colour microscopic cell morphology and staining and biochemistry descriptively according to Bergeys Manual of Determinative. Then the phosphate solubilization efficiency is measured by Pikovkayas medium. There are 37 from Talango and Poteran village that have potencial to be PSB and tend to get in 12 genera namely Bacillus Corynebacterium Staphylococcus Vibrio Aeromonas Pseudomonas Escherichia Edwardsiella Providencia Proteus Shigella and Yesrsinia. All of 37 isolate indicate that the average of phosphate solubilization efficiency level is 40 which of is Vibrio have the highest phosphate solubilizing efficiency level that is 232. This result assumed that the bacteria have potency to be biofertilzer.
