DIVERSITY AND POTENCY OF PHOSPHATE SOLUBILIZING MOLDS FROM POTERAN ISLAND SUMENEP MADURA

KEANEKARAGAMAN DAN POTENSI KAPANG PELARUT FOSFAT DARI PULAU POTERAN SUMENEP MADURA

ABSTRAK

Pulau Poteran Sumenep Madura memiliki tanah berbahan induk batu kapur dan mediteran merah menyebabkan tanah cenderung basa sehingga unsur fosfor terikat oleh unsur Ca. Salah satu kelompok mikroorganisme yang mampu mengubahnya adalah kapang. Penelitian ini dilakukan untuk mengetahui genus kapang pelarut fosfat dari Pulau Poteran dan potensinya menekan pertumbuhan kapang patogen tanah secara in vitro tanpa menyebabkan infeksi pada tembakau. Isolasi dilakukan di lima titik berbeda lalu diinkubasi di Laboratorium Biologi ITS. Isolat murni diuji dalam medium selektif Pikovskaya. Selanjutnya dilakukan dua uji potensi yaitu uji antagonis terhadap kapang patogen F. oxysporum serta uji patogenitas terhadap daun tembakau varietas Prancak 95. Hasil menunjukkan hanya empat isolat A B C D yang mampu membentuk zona bening diduga mendekati genus Aspergillus dengan rerata indeks kelarutan fosfat IKF tiap isolat A015 B013 C006 D008. Konsentrasi Ptersedia yang dihasilkan tiap isolat A927 B1024 C609 D421 ppm. Seluruh isolat diduga memiliki sifat antagonis dengan nilai penghambatan P untuk isolat A05065 B0435 C06305 D05393. Isolat A dan B diduga memiliki sifat patogen sedangkan isolat C dan D tidak menunjukkan gejala patogen pada daun tembakau.


ABSTRACT

Poteran Island Sumenep Madura has made from limestone soil and red mediterranean soil causing tend to be alkaline so the phosphorus element bound by Ca unsure. One group of microorganisms that are able to change it is mold. The aim of this research is to know the genera of phosphate solubilizing molds from Poteran Island and the potency to suppress of soil pathogeny mold without causing infection to tobacco under in vitro condition. Isolation conducted in five different spot then incubated in the laboratory of Biology ITS. Pure isolate tested by selective medium of Pikovskaya. Then conducted with two potency tests the antagonistic test toward pathogeny mold F. oxysporum and patogenicity test toward tobacco leaf Prancak 95 var. The result shows only four isolate A B C D that appeared the halozone form estimated to Aspergillus genera with the mean of soluble phosphorus index SPI of each isolate A015 B013 C006 D008. P-available concentration that resulted from each isolate A927 B1024 C609 D421 ppm. All isolates estimated have an antagonistic disposition with inhibitory I value for isolate A05065 B0435 C06305 D05393. Isolate A and B are estimated pathogen but isolate C and D are didnt shows any causes on tobacco leaf.
