ABSTRAK

Produksi enzim telah dilakukan melalui media fermentasi terendam dengan menggunakan jamur Aspergillus niger di dalam medium fermentasi yang mengandung dedak gandum sebagai substrat. Enzim yang dihasilkan antara lain enzim xylanase dalam jumlah besar dengan sedikit enzim selulase. Aspergillus niger ATCC 6275 di kultivasikan dalam erlenmeyer yang berisi substrat dedak gandum dan larutan medium fermentasi yang mengandung garam-garam mineral dan sumber nitrogen pada fermentasi terendam. Tujuan penelitian ini adalah menghasilkan enzim xylanase dengan aktivitas tinggi. Variabel yang digunakan adalah perbedaan konsentrasi substrat 1234 jumlah spora Ixl05-lxl06 lxl06-lxl07 1xl07-lxl08 sporaml medium fermentasi I dan II dan juga waktu inkubasi 2345 dan 6 hari. Dari hasil percobaan menunjukkan bahwa proses fermentasi terendam dengan medium larutan garam-garam mineral dan sumber nitrogen diperoleh aktivitas enzim xylanase terbesar adalah 2559 IUml pada konsentrasi substrat 1 jumlah spora lxl07-lxl08 sporaml dan waktu inkubasi 4 hari dengan konsentrasi protein yang terbentuk 08122 mgml. Enzim xylanase mempunyai aktivitas terbesar pada suhu 50 C dan pH 5. Purifikasi enzim dengan presipitasi ammonium sulfat menghasilkan aktifitas 3675 IUml. Sedangkan aktivitas enzim selulase 235 IUml dihasilkan pada konsentrasi substrat 1 jumlah spora Ixl07-lxl08 sporaml dan waktu inkubasi 3 hari. Pada aplikasi enzim terhadap pulp didapatkan persentase degradasi terbesar terhadap hemiselulosa selulosa dan lignin pada waktu reaksi 12 jam masing-masing sebesar 2469 selulosa 860 dan lignin 161.


ABSTRACT

Enzyme was produced under submerged fermentation by using Aspergillus niger fungi in the fermentation medium containing wheat bran as substrate. The xylanase enzyme has been produced from Aspergillus niger with a little amounts of cellulase. Aspergillus niger ATCC 6275 were cultivated in erlenmeyer containing wheat bran as substrate and fermentation medium containing minerals salt solutions and nitrogen source under submerged fermentation. The objective of the research is producing xylanase enzyme with high activity. The variable of this research was substrate concentration 1 2 3 and 4 spora concentration Ixl05-lxl06 lxl06-lxl 07 lxl07-lxl08 sporaml type of fermentation medium I and II and incubation time 2345 and 6 days. The experiment result showed that submerged fermentation process with medium containing mineral salt solution and nitrogen source gives the highest xylanase enzyme activity of 2559 IUml on 1 substrate concentration spora concentration Ixl07-lxl08 sporaml and 4 days incubation with protein concentration of 08122 mgml. Xylanase enzyme gives the highest activity at 50C and pH 5. Enzyme purification with ammonium sulphate precipitation gives xylanase activity of 3675 IUml. Cellulase enzyme with an activity of 235 IUml was produced on 1 substrate concentration spora concentration 1x10 -1x10 sporaml and 3 days incubation. The enzyme application into pulp gives the highest degradation of hemicellulose cellulose and lignin at 12 hours reaction that is 2469 860 and 161 respectively.
