GENUS AND HOPANOIDS ANALYSIS FROM PURE CULTURE SOIL BACTERIA OF PINE FOREST

ABSTRAK

Hopanoid merupakan senyawa triterpen pentasiklik yang terdapat pada bakteri. Distribusi senyawa hopanoid banyak ditemukan pada kelompok bakteri tanah. Senyawa hopanoid telah berhasil diidentifikasi dari kultur murni campuran bakteri tanah dan kemungkinan berasal dari kontribusi kelompok bakteri penyubur tanah. Pada penelitian ini dilakukan identifikasi senyawa hopanoid dari genus yang terkait isolat murni bakteri tanah hutan pinus. Hopanoid diperoleh dengan cara ekstraksi sel kering dengan kloroformmetanol 21 vv dan didegradasi dengan H5IO6NaBH4 KK fraksinasi asetilasi dengan asetatpiridin dan analisis menggunakan KG-MS. Penentuan genus bakteri tanah meliputi pewarnaan Gram pewarnaan kapsula pewarnaan endospora respirasi bakteri bentuk sel ukuran sel dan gerak bakteri. Selain itu digunakan analisa microbact system untuk penentuan genus. Hasil dari penelitian ini diidentifikasi 3 genus yaitu Clostridium Bacillus dan Klebsiella dari 4 isolat murni bakteri tanah hutan pinus. Masing-masing genus tersebut memproduksi senyawa hopanoid yang khas. 17H21H-homohop- 30-ol-31-asetat khas untuk Clostridium. Metil-hop-671721-dien dan 17H21H-metil hopan khas untuk Bacillus. Sedangkan metil-hop- 671721-dien 17H21H-bishomohop-30-ol-32-asetat 17H21H- metilhomohop-6-en-30-ol-31-asetat dan hop-1722en-35-asetat khas untuk Klebsiella. Kekhasan struktur hopanoid yang diperoleh memberikan indikasi bahwa senyawa hopanoid bisa digunakan sebagai marker keanekaragaman jenis bakteri penyubur tanah dalam suatu habitat tanah. Selain itu senyawa ini bisa digunakan sebagai indikator kesuburan tanah.


ABSTRACT

Hopanoids are pentacyclic triterpenoid lipids occurring in bacteria. Their distribution are widespread in soil bacteria and now well known. Hopanoids compound was successfully identified from mixed culture of soil bacteria. There is a possibility coming from contribution of bacteria which takes part in soil fertilizing. This reseach succesfully identified hopanoids and the type of genus that produced it from pure culture of pines forest. Hopanoids have been obtained from extraction freeze dried cells by chloroformmethanol 21 vv. Analysis of hopanoids using GC-MS toward hopanoids that is degradated by H5IO6NaBH4 fractionated by column Chromatography and acetylated by acetic acid piridin. The types of soil bacteria genus were characterized from Gram staining capsula staining endospore staining respiration cell type and motility. Analysis microbact culture is also used to identified bacteria genus. Three genus are Clostridium Bacillus and Klebsiella from pure culture of pines forest produced special hopanoids compound. 17H21H-methylhomohop-30-ol-31-OAc produced from Clostridium. Methyl-hop-671721-dien and 17H21H- metil hopan produced from Bacillus. Methyl-hop-671721-dien 17H21H-bishomohop-30-ol-32-OAc 17H21H-methylhomohop-6- en-30-ol-31-OAc and hop-1722en-35-OAc produced from Klebsiella. The special characteristics hopanoids compound from related genus can be used as indicator soils bacteria diversity. This compound can also be used as soil fertility indicator.
