ANALYSIS Of HOPANOID CONTENT FROM PLANTATION SOIL BACTERIA GROWING AFTER 48 HOURS AS SOIL FERTILITY INDICATOR

ABSTRAK

Hopanoid merupakan salah satu senyawa metabolit sekunder yang dihasilkan oleh bakteri. Analisis hopanoid pada bekteri tanah akan memberi gambaran hubungan antara keragaman bakteri tanah dengan kesuburan tanah.Bakteri yang diambil dari tanah kebun dibiakkan dalam cawan petri dan diahasilkan 19 koloni setelah inkubasi selama 48 jam. Hopanoid bakteri dapat diekstrak dari sel kering bakteri yang dipanen setelah 45 jam kemudian didegradasi dengan metode H5IO6NaBH4. Analisis KG MS digunakan untuk menentukan struktur hopanoid yang terdapat dalam bakteri. Jenis hopanoid yang dihasilkan oleh bakteri tanah adalah homohopan-6ene-31-Ol Asetat dengan berat sebesar 0.01 mgg sel kering. Asam lemak yang terdeteksi dapat berfungsi sebagai kontrol pada kemotaksonomi. Hasil analisis KG SM menunjukkan dihasilkannya tiga jenis asam lemak oleh bakteri tanah kebun yaitu asam behenat etil ester 003 mgg sel kering asam palmitat 001 mgg sel kering dan asam palmitat etil ester 002 mgg sel kering.


ABSTRACT

Hopanoid is one of secondary metabolites that produced by bacteria. Analysis hopanoid in soil bacteria could show the relation between soil fertility and bacteria diversity. Hopanoid from bacteria could be extracted from dried cell and then degraded with H5IO6NaBH4 method. Hopanoid structure had been predicted by GC MS method. The result shows that bacteria colonies harvested after 45 hours incubation produced homohopan-6ene-31-Ol Asetat 001mgg dried cells. In addition to hopanoid analysis fatty acids composition of such colonies had been also analysisby the same method.The result shows that bacteria colonies produced behenic acid ethyl ester 003 mgg dried cells palmititic acid 001 mgg dried cells dan palmitic acid ethyl ester 002 mg g dried cells. Analysis of fatty acids were used as control in chemotaxonomy.
